ACES, with its Chinese name N - (2-acetylamino) -2-aminoethanesulfonic acid and CAS number 7365-82-4, has an effective buffer range of pH 6.1-7.5 and a Pka value of 6.78 (25 ºC). ACES has weaker recombination ability with metal ions than ADA. Therefore, when using ACES in solutions containing metal ions, attention should also be paid to the stability constant. ACES absorbs 230nm ultraviolet light, which may interfere with spectrophotometry.
As a type of biological buffer, its primary application is undoubtedly as a pH stabilizer. Many biological reactions occur under neutral pH conditions, and the pH range of ACES is precisely between 6.1 and 7.5. It forms complexes with most metal ions, so it is commonly used in specific biochemical experiments. As ACES does not produce toxicity to cells, it can also be used in cell culture.
When identifying nucleic acids with larger molecular weights, ACES is more suitable than other buffering agents. As is well known, enzymes are sensitive to changes in pH values, and the vast majority of biochemical reactions require the catalytic action of specific enzymes to proceed. Therefore, in experiments that require enzymes, buffering agents are usually needed to stabilize the reaction environment of the experiment.
