PHARMACOKINETICS
Omeprazole was absorbed from small intestines after oral administration. The onset of the antisecretory effect of omeprazole occurred within one hour. Peak blood concentration of omeprazole was reached within 0.5 to 3.5 hours and the duration of inhibition lasted up to 24 hours. Omeprazole can be distributed in liver, kidney, duodenum and thyroid gland, and can pass through placenta easily. Usually, bioavailability is about 35% at a single dose and can be increased to 60 % at multiple doses. Protein binding is 95 % to 96 %. The plasma half-life is 0.5 to 1 hour in healthy subjects and is 3 hours in patients with chronic liver diseases. Omeprazole is metabolized by the cytochrome P450 oxidizing enzyme system of liver microsome in vivo, 80 % of metabolite was eliminated in urine, the remainder was excreted via bile and then eliminated in feces.
