Procaine hydrochloride is a local anesthetic that can temporarily block the conduction of nerve fibers and has anesthetic effects. It has strong effects, low toxicity, and is non addictive. However, it has weak penetration into the skin and mucous membranes and is not suitable for surface anesthesia. It is mainly used clinically for infiltration, spinal and conduction anesthesia.
The aqueous solution of this product is relatively stable between pH 3.0 and 5.0. Therefore, when preparing procaine hydrochloride injection, it is generally necessary to add acid to adjust the pH between 3.3 and 5.5, and strictly control the sterilization temperature and time. It is advisable to sterilize with circulating steam at 100 ºC for 30 minutes, and attention should be paid to light shielding and sealed storage. The aromatic primary amino group in the structure of this product reacts with sodium nitrite in dilute hydrochloric acid to form a diazonium salt, which then undergoes a coupling reaction with alkaline β - naphthol to produce a crimson colored azo compound.
Procaine hydrochloride is a white fine needle shaped crystal or crystalline powder at room temperature, odorless, slightly bitter and numbing in taste, easily soluble in water, soluble in ethanol, slightly soluble in chloroform, and almost insoluble in ether. The ester bonds in the structure of procaine hydrochloride can undergo hydrolysis to produce p-aminobenzoic acid and diethylaminoethanol. Under certain conditions, p-aminobenzoic acid can further decarboxylate to produce toxic aniline. Procaine hydrochloride contains aromatic primary amino groups in its structure, which are easily oxidized and discolored. pH, temperature, UV radiation, oxygen, and metal ions can all accelerate oxidation and discoloration.
