Introduction
The HiPure Stool DNA Kit is designed specifically for Stool DNA extraction.The kit is suitable for high purity extraction of microbial or host cell DNA from fecal samples ≤0.2g.The kit uses silica gel column purification technology and original inhibition factor adsorption technology.The adsorbent can adsorb humic acid and other inhibitors in the solution efficiently.The purified DNA can be directly used in PCR, Southern hybridization, enzyme digestion and other experiments.
Principle
HiPure silica column is based on a glass fiber filter membrane with high binding force.Under the condition of high concentration of ionizing agent (such as guanidine hydrochloride or guanidine isothiocyanate), the membrane can adsorb nucleic acid through physical factors such as hydrogen bonding and static electricity, while the protein or other impurities are not adsorted and removed.A membrane with adsorbed nucleic acids is washed to remove proteins and salts. Finally, the adsorbed nucleic acids can be eluted from the membrane with a low-salt buffer (such as sterilized water) or with water.The obtained nucleic acid has high purity and can be directly used in various downstream experiments. The fecal samples were homogenized in a lysate, and further lysed in a high-temperature water bath, releasing DNA into the lysate.The fecal impurities were removed by centrifugation, and the DNA was further purified through the column. Finally, the DNA was eluted by Buffer AE.The eluted DNA can be directly used in PCR, Southern hybridization, enzyme digestion and other experiments.

Storage conditions and Validity
Except for Buffer PCI and Proteinase K, the other components of the HiPure Stool DNA Kit can be dried and stored at room temperature (15-25ºC) for 12 months.It should be stored at 2-8ºC for long-term storage.Buffer PCI and Proteinase K must be stored at 2-8ºC.At low temperature, Buffer ATL may precipitate, which requires a 37ºC water bath to completely dissolve the precipitate.
