How to Prepare it?
The 10 x MOPSBuffer commonly used in electrophoresis buffer is prepared as follows:
(1) Using 41.8g MOPS and dissolving it into approximately 700mLDEPC treated water;
(2) Adjusting its pH to 7.0 using 2N NaOH;
(3) Add 20 mL of DEPC-treated 1MNaOAC and 20 mL of 0.5MEDTA (pH 8.0) to the solvent;
(4) Set the volume to 1L, filter through the impurities using a 0.45μm membrane and store the final extracted solvent at room temperature and away from light.
Because MOPS is chemically stable, the Pka value is stable and has no toxic non-permeable effect on organisms. It does not react chemically with many chemical test.It does not react chemically with many chemical agents and has no complexing ability with metal cations. It has many advantages as an amphoteric buffer and is therefore widely used in biosciences, medical diagnostics, etc.It is widely used in many fields such as biological science and medical diagnosis.
