Thrombin is derived from bovine plasma and has a molecular weight of 37KD. It is composed of two peptide chains (31KD and 6KD) through disulfide bonds. Thrombin is a protein hydrolytic enzyme formed after the activation of prothrombin (coagulation factor II). It catalyzes the hydrolysis of A and B peptides by fibrinogen, forming fibrin monomers. The monomers further aggregate and form blood clots with the participation of platelets, red blood cells, and white blood cells.
Due to its specificity in cleaving sequences and high hydrolysis efficiency, thrombin is widely used in the development of genetic engineering products. One of its applications is as a tool protease for specific cleavage of recombinant fusion proteins, particularly suitable for the biotechnology pharmaceutical industry, genetic engineering, biochemistry, molecular biology, and other research.
